Evaluation of Reagent Adding Methods of OCPC For the Determination of Serum Calcium
Abstract:Objective To evaluate two different methods of reagent adding of OCPC for the determination of serum calcium.Methods Method 1Mixing alcohol amine buffer(R1) with OCPC(R2)in the preparation tube and then putting the preparation reagent into sample tube(method 1 is the same as the manufacturer#39;s direction);method 2Adding R1 and R2 respectively and then mixing.Comparing the results of method 2 with results of automatic instrument and observation of precision and accuracy of this method.Results The VIS of EQC for serum calcium in our Laboratory was 0~289 when the assay of manufacturer#39;s direction was used;but PT of EQC for serum calcium has been 100 continuous since 2003 with method 2.Concentration of serum calcium of 30 samples was determined by OCPC method assays with Roche HITACHI MODULAR instrument and by the method 2 assays with MD100 semiautomatic instrument.It is highly correlative between the data obtained with the two methods(r=0.986.P<0.001).The rate of recovery was 99.5%.the CV with in run was 4.03% and the CVday to day was 5.6%.Conclusion The method 2 is simple,fast,accurate,avoiding interference,and suitable for clinical use.

我室引入全自动生化分析仪较晚,在相当长时间里一直用半自动生化分析仪测定血清钙。我们发现预先不配工作液,将两种试剂按顺序直接加入样品管中可以避免工作液管污染问题,效果也很好。现报告如下。
1 材料和方法
1.1 样本来源 标本来自本院患者;省室间质控物。
1.2 血清钙手工法测定 使用美国MD-100半自动生化分析仪,钙试剂盒和质控品均由中生北控生物科技股份有限公司提供,邻甲酚酞络合酮比色法(OCPC),最初我们完全按说明书操作,即预先配工作液,将乙醇胺缓冲液(R1)与邻甲酚酞络合酮(R2)等量混合,加到样品管中进行测定;后来改为R1与R2直接按顺序加入样品管中,混合均匀,预温5 min立即比色。
1.3 血清钙全自动生化分析仪测定 Roche HITACHI MODULAR型分析仪及原装试剂盒,OCPC法双试剂,质控两种:高值批号16366110;正常值批号16409804,按照公司规定的程序作检测。
1.4 统计学方法 计量资料以±s表示,对检测结果进行相关性分析。
